Breeding for microbiome-mediated disease resistance Organic Eprints

Plant-associated microbial communities play a crucial role for the expression of various plant traits including disease resistance. Increasing evidence suggests that host genotype influences the composition and function of certain microbial key groups, which, in turn, effects how the plant reacts to environmental stresses. Several studies indicate that modern plant breeding may have selected against plant traits essential for hosting and supporting beneficial microbes. However, they also highlight the presence of an exploitable genetic base for the regulation of the rhizosphere microbiota. Introduction In Switzerland, the organic sector has been trying to reduce the massive import of protein crops for feed by encouraging grain legume cultivation during the past years (Clerc et al, 2015). The main limiting factors of low yield stability and high weed pressure were successfully overcome through intercropping of peas and faba beans with barley and oats, respectively. Thus, the production of peas and faba beans grew continuously from 2010 to 2015 from 200t and 90t to 1170t and 560t, respectively (bioaktuell 2017). Modern plant breeding can no longer afford to ignore the interaction between plants and microbial key players. Increasing evidence suggests (i) that the expression of many plant traits (such as nutrient use efficiency or tolerances against biotic and abiotic stresses) is mediated by beneficial microorganisms and (ii) that there is an exploitable genetic base for the regulation of symbiotic relationships. Plant associated soil microbes are known to play an important role in the expression and stability of certain plant traits such as nutrient use efficiency and disease resistance. Arbuscular mycorrhizal fungi (AMF) form one of the primary mutualistic plant-microbe symbioses. Besides known benefits such as improved nutrient mobilisation (mainly phosphorus and zinc) and tolerance against abiotic stresses (mainly drought), an increasing number of studies highlight a significant role of AMF in the mediation of disease resistances and priming mechanisms. Introduction Mixed cropping, i.e. mixing different crops in the same field, provides agronomic advantages as increased productivity under low inputs conditions (e.g. for organic farming: Bedoussac et al. 2015) and higher yield stability (Raseduzzaman and Jensen 2017). In mixed cropping, choosing the right cultivars is critical for the performance of the mixture, as shown for pea-barley mixtures (Hauggaard-Nielsen and Jensen 2001) and maize-bean mixtures (Hoppe 2016). Ecological studies of the bio-inoculant Trichoderma hamatum LU592 in the root system of Pinus radiata Organic Eprints

The plant health- and growth-promoting biological inoculant (bio-inoculant) Trichoderma hamatum LU592 was transformed with the constitutively expressed green fluorescent protein (gfp) and hygromycin B resistance (hph) genes to specifically monitor the isolate in the root system of Pinus radiata within a strong indigenous Trichoderma population. A modified dilution plating technique was developed to allow the determination of the mycelia proportion of total propagule levels. LU592 was shown to colonize the rhizosphere most effectively when 105 spores per pot were applied compared with inoculum concentrations of 103 and 107 spores per pot. Ecological studies of Trichoderma bio-inoculants in the soil ecosystem of Pinus radiata Organic Eprints

Three isolates of different Trichoderma species were selected for this study. T. hamatum (LU592) and T. harzianum (LU686), were known to stimulate growth and improve establishment of P. radiata seedlings, and T. atroviride (LU132) had no stimulatory activity. The ability of these isolates to establish and survive in bulk soil, the rhizosphere, rhizoplane or endorhizosphere of P. radiata seedlings was determined. In addition, the effect of each isolate on P. radiata seedling development and growth was assessed and is currently being confirmed in a large scale commercial experiment.
